                  Development of ELISA Kit for Analysis of Hapten-Structured Toxin

ÖZET
          Mycotoxins are fungal secondary metabolites in hapten structure and produced by some fungus species

such as Aspergillus, Fusarium, Penicillium, and Alternaria. One of the most important food contaminants are
mycotoxins which is hazardous to human health. According to Food and Agricultural Organization 25% of agricultural
products are contaminated with mycotoxins worldwide. Mycotoxins, which are mostly heat tolerant small molecules,
cannot be destroyed by usual cooking procedures and hence cause severe health issues such as hepatotoxicity,
genotoxicity, oncogenicity, nephrotoxicity, immunomodulation, fertility and reproduction perturbation, central
nervous system damage or skin toxicity. Aflatoxin, one of the mycotoxins, has various types such as B1, B2, G1, G2,
M1 and M2. Aflatoxin B1 has been classified as group 1 carcinogen. The aim of this study is to develop an ELISA kit
for the detection of Aflatoxin B1 in contaminated foods. This ELISA kit will be developed using antibodies previously
developed in TUBITAK MAM.

          The main method to be used in this study is immunoassay, which is one of the most frequently used methods
in toxin detection. One of these methods is called competitive ELISA. Competitive ELISA is based on prevention of
the interaction between immobilized antigen/antibody and labelled antibody/antigen in the presence of the analyzed
molecule. Competitive ELISA tests can be applied in two ways. First, if the antibody is immobilized, antigen is labelled
and the labelled antigen is mixed with analyte. Secondly, if the antigen is immobilized, antibody is labelled and the
labelled antibody is mixed with analyte. In both ways, the signal detected in result of the interaction between
immobilized and labelled components of the assay decreases by increasing analyte concentration in the solution.

          A sensitive AFB1 ELISA kit developed in this study will be useful in terms of food safety.
